A simple method for gene walking using a gene-specific primer and a degenerate primer was described by Martin-Harris, et. al. (Analytical Biochemistry 399 (2010) 308–310). The method, termed sequential hybrid primer PCR (SHP-PCR), involves an initial PCR round using a gene-specific forward primer and a tagged reverse degenerate primer followed by two or three PCR rounds, each of which uses a gene-specific forward primer and a specially designed hybrid primer.
Monoquant supplies a set of primers comprising three hybrid primers and three tag primers which together enable the performance of an initial PCR followed by three sequential rounds involving three sequential hybrid primers. The quantity supplied is sufficient to enable 20 SHP-PCR studies to be performed.
